He gets a clue on the latter question during the ceremony, when the bride walks down the aisle. As soon as she passes Josuke's seat, and he sees the back of her dress, his eyes widen at the sight of her birthmark.
Okay... So he might know why he's here now.
Looking around though, he's pretty sure he doesn't recognize any of the other guests, so... Yeah. Still pretty confused. He keeps his confusion to himself until the ceremony is over, metaphorically sitting on his hands until the reception, at which point he tries to get a moment alone with the woman who he knows he must somehow be related to. That moment alone is hard to get, though, since she's always with her new wife (he can't really blame her for that), and since her stand keeps blocking him from trying to get closer (at least, he's assuming the stand is hers).
So there's Josuke, separated from his fiancée, trying to negotiate with Stone Free for an audience with its user. ]
No, please, you don't get it... I know the bride! I mean, I don't actually know her, but... We're cousins! Or... something. Anyway, can I please just talk to her for a minute??

Until Jolyne's quickly muttering something to Carly and stealing a quick kiss before standing and making her way over to him and the Stand.]
The fuck's this about us being cousins or something? Pretty damn sure my dad doesn't have any siblings. [She's assuming it's probably her dad's side of the family since he sure doesn't look remotely anything like her mom or anything.
She's leaning against the stalwart Stand a bit, a bit of string being recalled back to the Stand's hand that had been connected to Jolyne. That... is probably how she heard all of that.]
And don't bother asking me how I heard all that, let's just say string's pretty fuckin' useful. Now who the hell are you?
